Isoform 1 and isoform 2 show reduced expression in schizophrenic brain. Isoform 3 shows increased expression in schizophrenic and bipolar disorder brains while isoform 4 shows reduced expression.

Description:
The gamma-aminobutyric acid (GABA) A receptor is a multisubunit chloride channel that mediates the fastest inhibitory synaptic transmission in the central nervous system. This gene encodes GABA A receptor, beta 2 subunit. It is mapped to chromosome 5q34 in a cluster comprised of genes encoding alpha 1 and gamma 2 subunits of the GABA A receptor. Alternative splicing of this gene generates 2 transcript variants, differing by a 114 bp insertion.

Research Backgrounds

Function:

Ligand-gated chloride channel which is a component of the heteropentameric receptor for GABA, the major inhibitory neurotransmitter in the brain. Plays an important role in the formation of functional inhibitory GABAergic synapses in addition to mediating synaptic inhibition as a GABA-gated ion channel. The gamma2 subunit is necessary but not sufficient for a rapid formation of active synaptic contacts and the synaptogenic effect of this subunit is influenced by the type of alpha and beta subunits present in the receptor pentamer (By similarity). The alpha1/beta2/gamma2 receptor and the alpha2/beta2/gamma2 receptor exhibit synaptogenic activity. Functions also as histamine receptor and mediates cellular responses to histamine (By similarity).

PTMs:

Glycosylated.

Tissue Specificity:

Isoform 1 and isoform 2 show reduced expression in schizophrenic brain. Isoform 3 shows increased expression in schizophrenic and bipolar disorder brains while isoform 4 shows reduced expression.

Subunit Structure:

Heteropentamer, formed by a combination of alpha, beta, gamma, delta and rho chains. Interacts with UBQLN1 (By similarity). Interacts with KCTD8, KCTD12 and KCTD16; this interaction determines the pharmacology and kinetics of the receptor response, the KCTD proteins markedly accelerating the GABA-B response, although to different extents (By similarity). May interact with KIF21B (By similarity). Identified in a complex of 720 kDa composed of LHFPL4, NLGN2, GABRA1, GABRB2, GABRG2 and GABRB3 (By similarity).

Family&Domains:

The extracellular domain contributes to synaptic contact formation.

Belongs to the ligand-gated ion channel (TC 1.A.9) family. Gamma-aminobutyric acid receptor (TC 1.A.9.5) subfamily. GABRB2 sub-subfamily.

(A) The levels of GABAAR-α1-5, -β1-3, -γ1-3, -δ subunits, and D1R, D2R mRNA in the NAc were examined at the end of the behavioral tests. *p < 0.05 vs. control mice (n = 8 mice per group; two-way ANOVA). (B) Bar graphs show levels of GABAAR-α1, GABAAR-α2, GABAAR-β2, and GABAAR-β3 protein in the NAc normalized by the level of GAPDH were normalized by control levels. *p < 0.05 vs. control mice (n = 8 mice per group; two-way ANOVA). (C) Representative Western blots of biotin-labeled surface proteins (surface) and total proteins (total) D2R in the NAc. Na+-K+ ATPase served as internal control and GAPDH served as a negative control. Bar graphs indicate the ratio of proteins at the cell surface to their total levels. **p < 0.01 vs. control mice (n = 8 mice per group; Student’s t -test).
